应用案例一


Defective Uterine Spiral Artery Remodeling and Placental Senescence in a Pregnant Rat Model of Polycystic Ovary Syndrome

妊娠相关问题被认为与母体子宫螺旋动脉(SpA)重塑损伤有关,但其关联机制尚不清楚。多囊卵巢综合征的两种常见临床表现——高雄激素和胰岛素抵抗是否影响子宫SpA重塑也不清楚。该研究证实妊娠期间大鼠暴露于5-二氢睾酮(DHT)和胰岛素(INS)不仅会导致高雄激素症、胰岛素不耐受和更高的胎儿死亡率;还会使子宫和胎盘中血管生成相关基因表达失调,表现为:胎盘功能不全标志物和细胞衰老相关蛋白表达水平的显著增加。该研究对包含子宫、胎盘和胚胎的完整组织透明化及免疫标记,通过锘海LS18平铺光片显微镜进行全组织成像,CD31和α-平滑肌肌动蛋白(α-SMA)用于表征血管在子宫中的分布。结果表明:高雄激素血症和INS抵抗诱导母体子宫血管EC异常并减少SpA重塑。综上,该研究表明多囊卵巢综合征妊娠并发症的增加可能是由子宫SpA重塑、胎盘功能和胎盘衰老问题介导的。

Single-cell RNA sequencing reveals new subtypes of lens superficial tissue in humans

虽然研究人员通过单核RNA测序绘制了人眼前段细胞图谱,然而上皮细胞中是否存在晶状体干/祖细胞亚型以及人晶状体细胞分化的分子特征尚不清楚。该研究通过单细胞测序对18,596个人晶状体浅表组织细胞进行分析,发现两种特异性表达C8orf4和ADAMTSL4的晶状体上皮细胞亚型,一种新型纤维细胞亚型位于上皮附近;一种可能是晶状体上皮干细胞的ADAMTSL4+细胞亚群,并通过生物组织透明化技术锘海LS18平铺光片显微镜对完整晶状体进行成像,观察不同年龄人晶状体中CD24纤维细胞簇的空间分布,进一步分析了晶状体上皮细胞分化的轨迹。

半月板的血供状况决定半月板的恢复情况,是制定治疗方案的参考依据。本研究采用组织透明化方法对转基因小鼠双侧膝关节组织用锘海LS18平铺光片显微镜三维成像分析半月板血管内皮细胞,进而使用软件定量分析小鼠半月板血管分布,为今后对人类半月板的评价提供了方便,也为临床医生提供了更多的解剖学参考。

Spatiotemporal Dynamics of the Molecular Expression Pattern and Intercellular Interactions in the Glial Scar Response to Spinal Cord Injury

成年哺乳动物脊髓的神经再生能力较差,主要是由于缺乏内在神经元的再生和外源性因素——损伤引起的神经胶质疤痕,抑制或促进再生。本研究利用空间转录组学(ST)构建了脊髓损伤后瘢痕形成过程中小鼠脊髓组织范围内基因表达模式,并局部分析了疤痕区域从前缘到核心的基因表达梯度,以进一步了解疤痕微环境。并通过组织透明化锘海LS18平铺光片显微镜对小鼠脊髓损伤后形成的疤痕组织进行三维可视化,结果表明:7 dpi时创面层粘连蛋白(Laminin)高度富集,纤维连接蛋白(Fn1)和胶原蛋白(Collagen IV)的表达相对较低。疤痕的空间异质性证实了以前关于疤痕结构的概念,为疤痕形成提供了一些新的线索,并为中枢神经系统损伤的治疗提供了宝贵的资源

Three-dimensional topography of eye-specific domains in the lateral geniculate nucleus of pigmented and albino rats

正常大鼠初级视觉皮层(V1)中存在眼优势柱(ODCs),且外侧膝状体背核 (dLGN)的同侧眼区域分离成少数斑块。为了研究dLGN特异性斑块的三维拓扑结构及其与ODCs的关系,研究人员在左右眼注射带有不同荧光标记的顺行示踪剂,通过组织透明化锘海LS18平铺光片显微镜对LGN进行整体成像,可视化LGN的三维形态,该研究发现从任何角度观察dLGN都呈现为网格状结构,并且在幼鼠睁眼前后发育完成,异常视觉经验影响但不中断网格结构的发育。在白化Wistar大鼠中,dLGN同侧眼区域斑块较少,特别是在中央视野附近。该研究为dLGN同侧斑块如何产生提供了新见解,并揭示了啮齿类与灵长类动物外侧膝状体背核拓扑结构的差异。
